New organic compounds as mediators and polymers as supports for enzyme immobilization have been investigated and applied for the bioelectroanalysis of biologicaly important compounds in carbon-based electrochemical systems. Bioconversion rate and efficiency of the glycerol-3-phosphate in reactor containing immobilized glycerol phosphate oxidase were investigated. Mathematical models of few heterogeneous bioelectrocatalytic systems have been created.
The kinetics of oxidation of artificial substrates catalyzed by novel fungal laccases was investigated. The influence of buffer solution components, pH and inhibitor action of halides on the laccases activity was evaluated. The function of the laccases on gold electrodes containing naked gold nanoparticles as well as modified by specific thiol compounds were analyzed.
Small angle X-ray diffraction, resonance Raman spectroscopy with 413 nm excitation, and non-resonance Raman technique with 785 nm excitation were used to study the influence of entrapped cytochrome c on the structure of hydrated phytantriol liquid-crystalline phases as well as conformational changes of heme group and secondary structure of the protein.
The self-organization of bioluminescent E. coli in suspensions was investigated. The experimental and numerical results showed that luminous bacteria form a weak gel near interfaces.

The electrochemical impedance spectra (EIS) of tethered bilayer membranes (tBLMs) were analyzed, and the analytical solution for the spectral response of membranes containing natural or artificially introduced defects was derived. The analysis carried out shows that the EIS features of an individual membrane defect cannot be modelled by conventional electrical elements. The primary reason for this is a complex nature of an impedance of the submembrane ionic reservoir separating the phospholipid layer and the solid support. We demonstrate that its EIS response, in case of radially symmetric defects, is described by the Hankel functions of a complex variable. Therefore, neither the impedance of the submembrane reservoir, nor the total impedance of tBLMs can be modelled using the conventional elements of the equivalent electrical circuits of interfaces. The presented analysis provides practical tools to evaluate the defect density in tBLMs, which could be utilized in tBLM-based biosensor applications. Alternatively, if the parameters of the defects, e.g., ion channels, such as the diameter and the conductance are known, the EIS data analysis provides possibility to estimate other physical parameters of the system, such as thickness of the submembrane reservoir, and its conductance. Finally, current analysis demonstrates possibility to discriminate between the situations, in which the membrane defects are evenly distributed or clustered on the surface of tBLMs. Such sensitivity of EIS could be used for elucidation of the mechanisms of interaction between the proteins and membranes. Surface-enhanced Raman spectroscopy (SERS) was used to detect different spectral features of small (1-2 nm) and large (5-10 nm) synthetic amyloid Ab-42 oligomers, exhibiting high and no detectable neurotoxicity, respectively. Adsorption of peptides at self-assembled monolayers (SAM) terminated by methyl and pyridinium groups was employed to differentiate toxic and non-toxic oligomers. Three SAMs were analyzed: hydrophobic heptanethiol (HT) and octadecanethiol (ODT) as well as positively charged N-(6-mercapto)hexylpyridinium (MHP) chloride. SERS study revealed two-fold adsorption effect, changes in the monolayer structure and appearance of new bands associated with the adsorbed peptides. A band at 1387 cm-1, observed as a result of the SAM and Ab-42 interaction, is tentatively assigned to the peptide symmetric stretching vibration of carboxylate groups, and appears to be the most prominent spectral feature distinguishing toxic oligomers from the non-toxic Ab-42 forms. This band was identified in the spectra of Ab-42 adsorption on heptanethiol and MHP monolayers, while no clear perturbations were observed in the case of ODT monolayer.

Interaction of 2,5-cyclohexadien-1,4-dione (p-benzoquinone) with o-, m-, p-substituted aniline derivatives was investigated. p-Benzoquinone reacted with aqueous solutions of various amines rather easily, and 2-arylamino-1,4-benzoquinones were formed. It was established that the yield and purity of 2-substituted benzoquinones depended on the reaction time, temperature, dilution and the ratio of reactants. The course of reactions was greatly influenced by the nature of substituents in the aromatic ring of amines. The synthesized 2-arylamino-cyclohexa-2,5-diene-1,4-diones were tested as new materials for electrochemical biocatalysis.
The synthesis of dodecaethylene glycol from dimezyltetraethylene glycol and an excess of tetraethylene glycol was studied. It was established that the impurities of this reaction were difficult to separate from the target product and their amount greatly depended on the reaction temperature, time, dilution, and stirring intensity.
Continuing our search for methods of synthesis of fused 1,5-benzodiazepine derivatives, we studied the transition metal induced reductive N-heterocyclization of 5-acyl-1-(2-nitrobenzoyl)-1,3,4,5-tetrahydro-2H-1,5-benzodiazepin-2-ones and synthesized derivatives carrying 6,7-dihydroquinazolino[3,2-a][1,5]benzodiazepine fragment. Quantum-chemical reactivity descriptors were calculated to explain the peculiarities of reductive cyclization.

We have examined the voltammetric characteristics of neuromodulating agent 6,7-dinitroquinoxa-line-2,3-dione (DNQX), its 6-mononitro derivative, and quinoxaline-2,3-dione, their reactivity towards single-electron transferring flavoenzymes NADPH:cytochrome P-450 reductase and NADPH:adrenodoxin reductase/adrenodoxin complex, their reactivity towards a two-electron transferring flavoenzyme DT-diaphorase, and their cytotoxicity in bovine leukaemia virus-tarnsformed lamb kidney fibroblasts (line FLK). The cytotoxicity of the above nitroheterocyclic compounds was prooxidant, i.e., having been prevented by the antioxidants, and that it correlated with the rates of their single-electron enzymatic reduction and voltammetric reduction potentials, which were similar to those of o-dinitrobenzene and p-nitroacetophenone, respectively. The reactions catalyzed by DT-diaphoresis exerted negligible role in the cytotoxicity of compounds. The cytotoxicity of quinoxaline-2,3-dione was much lower, this compound was inactive towards above mentioned flavoenzymes. Thus, our data characterize the mode of cytotoxicity of DNQX, which may be important as a side-effect of this drug.

Investigation of the Escherichia Coli Endoribonuclease G Activity Changes during Infection of Bacteriophage T4. Dr. L. Truncait臈. 2011鈥2015.
A sequence specific RegB endoribonuclease encoded by T4 related bacteriophages is usually toxic to the Escherichia coli cells when cloned and expressed in high quantities. In this study we cloned into expression vectors regB genes from different T4 like bacteriophages, which infect either E. coli (phages RB69, RB49, vB_EcoM_VR7) or Aeromonas salmonicida (phage 44RR). Recombinant endoribonucleases were tested for their activities in vivo by analysing their own mRNAs by primer extension analysis. All RegB proteins were active and showed high toxicity to the E. coli cells after induction. However, after mutagenesis of cloned regB genes we were able to express RegB proteins, which were less toxic to the cells.
Screening and Identification of Coumarins-Degrading Microorganisms. Application of NAD- And PQQ-Dependent Alcohol Dehydrogenases for Bioconversion. Dr. R. Me拧kys. 2011鈥2014.
Rhodococcus sp. K5 capable of growth on coumarin and melilotic acid was isolated for the first time. The coumarin inducible proteins were identified by SDS-PAGE and de novo protein sequencing. Quinohemoprotein alcohol dehydrogenases (ADHs) are attractive catalysts because of their wide substrate specificity and non-diffusible cofactor. ADH IIG from Pseudomonas putida HK5 was capable to oxidize various primary or secondary aliphatic and cyclic amino alcohols stereo selectively. An optimal pH range for ADH IIG activity was shifted to more acidic side (pH 4鈥6) when amino alcohols were used as substrates. The ADH IIG covalently immobilized on silica gel by cross-linking with glutaraldehyde or 1-ethyl-3(3-dimethylaminopropyl) carbodiimide exhibited a higher storage stability and thermo stability as well as an improved activity in the presence of organic solvents comparing with the free enzyme. The affinity of ADH IIG to substrates was not significantly changed in the presence of organic solvents or after the immobilization of the enzyme.

To Elucidate of RasGAP and Nck1 Interaction Mechanisms. Dr. M. Valius.
Previously we have shown that Ras GTPase activating protein (RasGAP) constantly interacts with adaptor protein Nck1; this interaction increases RasGAP ability to inactivate Ras. Now we show that this interaction can take place only in the cells that have contact with the substrate. Detachment of cells from the substrate leads to dissipation of RasGAP-Nck complex, down-regulation of RasGAP activity and increase of active Ras species in the suspended cells. The data provide mechanistic explanation how the attachment of adherent cells to the substrate increases their Ras activity, which serves as anti-apoptotic factor and increases attached cell survival.

The aim was to study the effect of histone deacetylase inhibitor EGCG on differentiation and apoptosis of human cancer cells.
Epigenetic changes such as histone acetylation and DNA methylation play an important role in human cancer cells. Chemical agents that affect histone deacetylases and DNA methyltransferases could be potential targets in cancer treatment. In this work it was found out that natural DNA methyltransferases inhibitor EGCG reduces cell proliferation and increases cell death 鈥 apoptosis, in both NB4 leukaemia and HeLa cervical cancer cells. We have shown by flow cytometry method that EGCG and its combination with retinoic acid induce differentiation in NB4 cells.

JNK Signalling after the Treatment with Natural Toxins in the Kidney-Derived Cells. Dr. V. Bukelskien臈.
Natural toxins referred to as mycotoxins producing mould species are extremely common. They can grow on a wide range of substrates under environmental conditions, and are linked to a variety of acute and chronic adverse health effects. In our study, effect of mycotoxins deoxynivalenol (DON) and ochratoxin A (OTA) were studied in model kidney-derived cells (BHK21). The results were compared with processes in model liver cell line (MH-22A). We have found that sensitivity to OTA and DON of both kinds of cells was similar and rather small in comparison to other kinds of cells. After 24 h treatment with tested mycotoxins, we observed the activation of c-Jun N-terminal kinase (JNK) in MH-22A cell culture, but this effect was inappreciable in BHK21 cell culture. However, we have registered the antiapoptotic role of JNK in both kidney- and liver-derived cells. Investigating the role of c-Jun, which is the main substrate of JNK, we have found that OTA and DON induced c-jun expression which correlated with proapoptotic effect of c-Jun.
